Exosome

Exosomes are membrane-bound extracellular vesicles (EVs) in size from 30 to 150 nanometers, which are generated in the endosomal compartment of most eukaryotic cells. Exosomes are extracellular vesicles with a unique biogenesis pathway through multivesicular bodies. Exosome formation starts with the invagination of the multivesicular bodies or late endosomes, then generates intraluminal vesicles. MVBs are trafficked to the internal side of the plasma membrane once formed. Exosomes contain different cargoes: proteins, lipids, and nucleic acids.

Exosomes Therapy

Exosomes are applied to analyze and determine the patient diagnosis, prognosis, progress and chemoresistance status, especially for cancer detection in blood samples. Comparing to traditional biomarker detection methods, exosomes for disease diagnosis has advantages of high sensitivity, high specificity, non-invasive.

Demonstration studies and researches show that exosomes as drug carriers can improve drug inhibition and bioavailability, reduce drug side effects. Exosomes in cancer treatment have 3 strategies:

  • Drug carrier to encapsulate drugs
  • Action as bio-active molecules to regulate tumor micro-environment
  • Inhibit tumor growth and metastasis

Therapeutic Exosomes

Exosomes also can treat autoimmune conditions as multiple sclerosis and rheumatoid arthritis. Anti-inflammatory agents are carried to regulate immune responses.

Exosome
  • Exosomes released from oral keratinocytes can accelerate wound healing.
  • Exosome shows a safe and effective treatment for low back pain.
  • Exosome-based vaccines have potential in the generation of strong cytotoxic T-lymphocyte for colorectal cancer.
  • Exosomes are excellent carriers for chemotherapeutic agents(doxorubicin, paclitaxel), engineered exosomes deliver drugs directly tumors, and minimize off-target effects.
  • Exosomes has the ability to cross biological barriers, like blood-brain barrier, this makes it attractive for neurological disease treatments like glioblastoma and Alzheimer’s disease.
  • Exosomes from mesenchymal stem cells (MSCs) promote tissue repair, particularly in cardiac repair post-myocardial infarction.
